Bortezomib plus melphalan and prednisone for initial treatment of multiple myeloma (Velcade as Initial Standard Therapy in Multiple Myeloma)

Phase 3N = 682NCT00111319

Population
Previously untreated patients with symptomatic myeloma who were ineligible for high-dose therapy.
Design
Phase 3 trial in 682 previously untreated patients with myeloma ineligible for high-dose therapy, randomly assigned to nine 6-week cycles of melphalan 9 mg/m2 and prednisone 60 mg/m2 on days 1 to 4, alone or with bortezomib 1.3 mg/m2 (days 1, 4, 8, 11, 22, 25, 29 and 32 in cycles 1 to 4; days 1, 8, 22 and 29 in cycles 5 to 9). The primary end point was time to disease progression.
Primary endpoint
TTP
Arms
  • Bortezomib, melphalan, prednisone (VMP) (experimental, n = 344): Melphalan 9 mg/m2 and prednisone 60 mg/m2 days 1-4 plus bortezomib 1.3 mg/m2, nine 6-week cycles
  • Melphalan, prednisone (MP) (control, n = 338): Melphalan 9 mg/m2 and prednisone 60 mg/m2 days 1-4, nine 6-week cycles
