Combined etoposide, ifosfamide, and cisplatin in the treatment of patients with advanced thymoma and thymic carcinoma: an intergroup trial

N = 34

Population
34 patients with advanced thymoma or thymic carcinoma entered; 28 evaluable after pathology review.
Design
Intergroup trial, July 1995 to February 1997: 34 patients with advanced thymoma or thymic carcinoma entered; etoposide 75 mg/m2, ifosfamide 1.2 g/m2 and cisplatin 20 mg/m2, each on days 1 to 4, every 3 weeks for four cycles (VIP). Pathology review excluded 6 patients, leaving 28 evaluable. The abstract does not name a phase or response criteria.
Arms
  • VIP (experimental, n = 34): Etoposide 75 mg/m2, ifosfamide 1.2 g/m2 and cisplatin 20 mg/m2 on days 1 to 4, every 3 weeks for four cycles

ORR28 evaluable patients (pathology review excluded 6 of 34)finalmedian follow-up 43 monthsLoehrer 200132% (16–52%)—No complete responses and 9 partial responses. Median duration of response 11.9 months (range under 1 to 26); median follow-up 43 months (range 12.8 to 52.3). Toxicity was predominantly myelosuppression. The authors conclude that VIP has moderate activity and that, with limited follow-up, the results appear inferior to other regimens reported in large phase II trials.
OS28 evaluable patients (pathology review excluded 6 of 34)finalmedian follow-up 43 monthsLoehrer 2001median 31.6 months; 89% at 12 mo; 70% at 2 y—Median overall survival 31.6 months; Kaplan-Meier 1-year and 2-year survival rates of 89% and 70%. No confidence intervals are printed.
