SWOG S0777

Bortezomib with lenalidomide and dexamethasone versus lenalidomide and dexamethasone alone in patients with newly diagnosed myeloma without intent for immediate autologous stem-cell transplant (SWOG S0777): a randomised, open-label, phase 3 trial

Phase 3N = 525NCT00644228

Population
Patients aged 18 years and older with newly diagnosed multiple myeloma without intent for immediate autologous stem-cell transplant (randomisation stratified by intent to transplant).
Design
Randomised (1:1), open-label phase 3 trial in patients aged 18 years and older with newly diagnosed myeloma, CRAB criteria and measurable disease, ECOG performance status 0-3, from SWOG and NCTN institutions; randomisation stratified by International Staging System stage and intent to transplant. VRd: eight 21-day cycles of bortezomib 1.3 mg/m2 intravenously on days 1, 4, 8 and 11, lenalidomide 25 mg on days 1-14 and dexamethasone 20 mg on days 1, 2, 4, 5, 8, 9, 11 and 12. Rd: six 28-day cycles of lenalidomide 25 mg on days 1-21 and dexamethasone 40 mg on days 1, 8, 15 and 22. Primary endpoint progression-free survival by a prespecified one-sided stratified log-rank test at a significance level of 0.02; analyses were intention to treat.
