Association of measurable residual disease with survival outcomes in patients with acute myeloid leukemia: a systematic review and meta-analysis

Phase meta-analysisN = 11,151

Population
Patients with AML in 81 publications (11,151 patients); studies reporting only outcomes after hematopoietic cell transplantation were excluded.
Design
Systematic review (PubMed, Embase, MEDLINE; studies published January 2000 to October 2018) with Bayesian hierarchical meta-analysis of survival probabilities and hazard ratios by MRD status for disease-free and overall survival. 81 publications.
Arms
  • MRD negative (experimental): Achieved MRD negativity
  • MRD positive (control): MRD present

OSAll pooled studies (81 publications)primaryShort 202068% at 5 y34% at 5 yHR 0.36Hazard ratio for achieving MRD negativity; the interval published is a 95% Bayesian credible interval of 0.33 to 0.39. The abstract gives the estimated overall survival (68% without MRD, 34% with MRD) in the same sentence as the 5-year disease-free survival; the open full text states the 5-year estimates for OS as 68% (95% CrI 63-73) and 34% (28-40).
