PRIME

A Randomized, Multicenter, Phase 3 Study to Compare the Efficacy of Panitumumab in Combination With Oxaliplatin/ 5-fluorouracil/ Leucovorin to the Efficacy of Oxaliplatin/ 5-fluorouracil/ Leucovorin Alone in Patients With Previously Untreated Metastatic Colorectal Cancer

Phase 3N = 1,183NCT00364013

Population
Previously untreated metastatic colorectal cancer
Design
Multicentre randomised phase 3 trial (1:1) of panitumumab plus FOLFOX4 versus FOLFOX4 alone as first-line treatment of metastatic colorectal cancer (ECOG performance status 0 to 2, tissue available for biomarker testing). The primary endpoint was progression-free survival and overall survival was secondary. Results were prospectively analysed by tumour KRAS status; KRAS results were available for 93% of the 1,183 patients randomly assigned. The 2013 report is a prospective-retrospective analysis by RAS (KRAS or NRAS) and BRAF status.

Arms
  • FOLFOX4 (control): FOLFOX4 (infusional fluorouracil, leucovorin, oxaliplatin)
  • Panitumumab plus FOLFOX4 (experimental): Panitumumab plus FOLFOX4
