PLCO

Prostate, Lung, Colorectal, and Ovarian (PLCO) Cancer Screening Trial: prostate-cancer screening component

N = 76,693NCT00002540

Population
Men aged 55-74 years at entry (full text of the 2009 report), randomised at 10 US study centres from 1993 through 2001.
Design
Randomised screening trial at 10 US study centres (men randomised 1993 through 2001). Men in the screening group were offered annual PSA testing for 6 years and digital rectal examination for 4 years; participants and their health care providers received the results and decided on follow-up evaluation. The control group received usual care, which sometimes included screening.
Primary endpoint
Prostate cancer mortality
Arms
  • Annual screening (experimental, n = 38,343): Annual PSA testing for 6 years and digital rectal examination for 4 years
  • Usual care (control, n = 38,350): Usual care, which sometimes included screening

Estimated yearly screening-phase PSA testing was 84% in the intervention arm and 46% in the control arm. In the same report, deaths from prostate cancer were 255 (intervention) and 244 (control), rate ratio 1.04 (95% CI 0.87-1.24); median follow-up 14.8 years (intervention) and 14.7 years (control). Arm sizes in the 2017 report differ slightly from the 2009 report's.
