Olaratumab and doxorubicin versus doxorubicin alone for treatment of soft-tissue sarcoma: an open-label phase 1b and randomised phase 2 trial
Phase 1/2N = 148NCT01185964
- Population
- 148 patients (15 in phase 1b; 133 randomised in phase 2) with locally advanced or metastatic soft-tissue sarcoma not previously treated with an anthracycline
- Design
- Open-label phase 1b (15 patients treated with olaratumab plus doxorubicin) and randomised phase 2 (1:1, minimisation) at 16 US sites; anthracycline-naive patients with advanced or metastatic soft-tissue sarcoma, ECOG performance status 0 to 2 and tissue available for PDGFR-alpha testing. Up to eight 21-day cycles. Phase 2 primary endpoint PFS with two-sided alpha 0.2 and power 0.8.

- Arms
- Doxorubicin (control, n = 67): Doxorubicin 75 mg/m2 on day 1 of each 21-day cycle, up to eight cycles
- Olaratumab plus doxorubicin (experimental, n = 66): Olaratumab 15 mg/kg on days 1 and 8 plus doxorubicin 75 mg/m2 on day 1 of each 21-day cycle, up to eight cycles

- Olaratumab plus doxorubicin (experimental)
- Doxorubicin (control)
Median-only curves assume a constant hazard (exponential) and fit plateaus, as seen with some immunotherapies, poorly.
