Perioperative chemotherapy versus surgery alone for resectable gastroesophageal cancer

Phase 3

Population
Patients with resectable adenocarcinoma of the stomach, oesophagogastric junction or lower oesophagus.
Design
Randomised comparison of perioperative chemotherapy plus surgery (250 patients) with surgery alone (253 patients). Chemotherapy: three preoperative and three postoperative cycles of epirubicin 50 mg/m2 and cisplatin 60 mg/m2 on day 1 with fluorouracil 200 mg/m2/day by continuous infusion for 21 days (ECF). Registered as ISRCTN93793971 (Current Controlled Trials); no ClinicalTrials.gov number is recorded.

Arms
  • Surgery alone (control, n = 253): Surgery with no perioperative chemotherapy
  • Perioperative ECF plus surgery (experimental, n = 250): Three cycles of epirubicin, cisplatin and infused fluorouracil before and three after surgery
