IESS-I

Multimodal therapy for the management of primary, nonmetastatic Ewing's sarcoma of bone: an Intergroup Study

N = 342

Population
Previously untreated eligible children with primary nonmetastatic Ewing's sarcoma of bone; 342 entered
Design
Randomised; group I institutions randomised patients between treatment 1 (radiotherapy to the primary lesion plus VAC, i.e. cyclophosphamide, vincristine, dactinomycin, plus doxorubicin) and treatment 2 (VAC without doxorubicin); group II institutions randomised between treatment 2 and treatment 3 (VAC plus bilateral pulmonary radiotherapy). 342 eligible previously untreated children entered between May 1973 and November 1978 according to the 1990 long-term follow-up report; the original 1981 report describes 264 randomised children and adults treated at 83 institutions of three study groups from 1973 to 1978 (the abstracts do not explain the difference).
Arms
  • Treatment 2: VAC (control): Radiotherapy to the primary lesion plus cyclophosphamide, vincristine and dactinomycin (VAC)
  • Treatment 1: VAC plus doxorubicin (experimental): Radiotherapy to the primary lesion plus VAC and doxorubicin (ADR)
  • Treatment 3: VAC plus bilateral pulmonary radiotherapy (experimental): Radiotherapy to the primary lesion plus VAC and bilateral pulmonary radiotherapy (BPR)
