Idelalisib and rituximab in relapsed chronic lymphocytic leukemia

Phase 3N = 220NCT01539512

Population
Relapsed CLL with decreased renal function, previous therapy-induced myelosuppression, or major coexisting illnesses (patients less able to undergo standard chemotherapy)
Design
Multicentre, randomised, double-blind, placebo-controlled phase 3 study; 220 patients randomly assigned to rituximab plus idelalisib 150 mg twice daily or rituximab plus placebo. Primary endpoint PFS. At the first prespecified interim analysis the study was stopped early on the recommendation of the data and safety monitoring board owing to overwhelming efficacy.

Median-only curves assume a constant hazard (exponential) and fit plateaus, as seen with some immunotherapies, poorly.
