HD21
HD21 for Advanced Stages Treatment Optimization Trial in the First-line Treatment of Advanced Stage Hodgkin Lymphoma; Comparison of 6 Cycles of Escalated BEACOPP With 6 Cycles of BrECADD
Phase 3N = 1,500NCT02661503
- Population
- Adults aged 60 years or younger with newly diagnosed advanced-stage classic Hodgkin lymphoma (Ann Arbor stage III or IV, or stage II with B symptoms and large mediastinal mass and/or extranodal lesions); median age 31 years.
- Design
- Randomised, multicentre, parallel, open-label phase 3 trial at 233 sites in nine countries: 4 or 6 cycles (21-day intervals) of BrECADD or escalated BEACOPP, guided by PET after two cycles. Hierarchical co-primary objectives: lower treatment-related morbidity, then non-inferior progression-free survival with an absolute margin of 6 percentage points; superiority of PFS tested if non-inferiority was shown. Intention-to-treat analysis; 1500 patients enrolled and 1482 analysed.
- Primary endpoint
- TRMB, PFS
- Arms
- BrECADD (experimental, n = 749): Brentuximab vedotin, etoposide, cyclophosphamide, doxorubicin, dacarbazine and dexamethasone, 4 or 6 cycles guided by PET after two cycles
- eBEACOPP (control, n = 751): Escalated bleomycin, etoposide, doxorubicin, cyclophosphamide, vincristine, procarbazine and prednisone, 4 or 6 cycles guided by PET after two cycles
- People
- Borchmann P
