Adjuvant ipilimumab versus placebo after complete resection of high-risk stage III melanoma

Phase 3N = 951NCT00636168

Population
Patients with completely resected stage III cutaneous melanoma (excluding lymph node metastasis of 1 mm or less or in-transit metastasis) who had not received previous systemic therapy for melanoma
Design
Double-blind, randomised (1:1), placebo-controlled phase 3 trial. Ipilimumab 10 mg/kg or placebo every 3 weeks for four doses, then every 3 months for up to 3 years. Randomisation stratified by disease stage and geographical region. Primary endpoint recurrence-free survival assessed by an independent review committee, analysed by intention to treat; secondary endpoints included overall survival and distant metastasis-free survival.
Primary endpoint

Arms
  • Ipilimumab 10 mg/kg (experimental, n = 475): Ipilimumab 10 mg/kg every 3 weeks for four doses, then every 3 months for up to 3 years
  • Placebo (control, n = 476): Placebo on the same schedule

Median-only curves assume a constant hazard (exponential) and fit plateaus, as seen with some immunotherapies, poorly.
