EORTC 1325/KEYNOTE-054

Pembrolizumab versus placebo after complete resection of high-risk stage III melanoma

Phase 3N = 1,019NCT02362594

Population
Completely resected AJCC-7 stage IIIA (at least one lymph node metastasis larger than 1 mm), IIIB or IIIC (without in-transit metastasis) cutaneous melanoma
Design
Double-blind, randomised (1:1), placebo-controlled phase 3 trial. Pembrolizumab 200 mg or placebo every 3 weeks for 18 doses (about 1 year) or until recurrence or unacceptable toxicity. Coprimary endpoints: recurrence-free survival in the overall ITT population and in the PD-L1-positive subgroup.
Primary endpoint
RFS
Arms
  • Pembrolizumab (experimental, n = 514): Pembrolizumab 200 mg intravenously every 3 weeks, 18 doses
  • Placebo (control, n = 505): Placebo intravenously every 3 weeks, 18 doses
