ECHELON-1

A Randomized, Open-label, Phase 3 Trial of A+AVD Versus ABVD as Frontline Therapy in Patients With Advanced Classical Hodgkin Lymphoma

Phase 3N = 1,334NCT01712490

Population
Previously untreated stage III or IV classic Hodgkin lymphoma; 664 assigned to A+AVD and 670 to ABVD.
Design
Open-label, multicentre, randomised phase 3 trial of brentuximab vedotin plus doxorubicin, vinblastine and dacarbazine (A+AVD) versus doxorubicin, bleomycin, vinblastine and dacarbazine (ABVD), up to six cycles, in previously untreated stage III or IV classic Hodgkin lymphoma. Primary endpoint: modified progression-free survival as adjudicated by an independent review committee; key secondary endpoint: overall survival.
Primary endpoint
mPFS
Arms
  • A+AVD (experimental, n = 664): Brentuximab vedotin plus doxorubicin, vinblastine and dacarbazine, up to six cycles
  • ABVD (control, n = 670): Doxorubicin, bleomycin, vinblastine and dacarbazine, up to six cycles
