COMPARZ

Pazopanib Versus Sunitinib in the Treatment of Locally Advanced and/or Metastatic Renal Cell Carcinoma

Phase 3N = 1,110NCT00720941

Population
First-line clear-cell metastatic renal cell carcinoma
Design
Randomised 1:1 phase 3 non-inferiority trial of pazopanib 800 mg once daily (continuous) versus sunitinib 50 mg once daily for 4 weeks followed by 2 weeks off, in 6-week cycles, as first-line therapy. Primary end point progression-free survival by independent review, powered to show non-inferiority of pazopanib (predefined margin: upper bound of the 95% confidence interval below 1.25). Secondary end points included overall survival, safety and quality of life.
Primary endpoint
PFS
Arms
  • Pazopanib (experimental, n = 557): Pazopanib 800 mg orally once daily, continuous
  • Sunitinib (control, n = 553): Sunitinib 50 mg orally once daily, 4 weeks on and 2 weeks off, in 6-week cycles
