Cytosine arabinoside with daunorubicin or adriamycin for therapy of acute myelocytic leukemia: a CALGB study

Population
Patients with acute myelocytic leukemia, reported in two age groups: younger than 60 years and older than 60 years.
Design
Randomised comparison, as described in the abstract, of daunorubicin 30 mg/m2, daunorubicin 45 mg/m2 or adriamycin 30 mg/m2 given on days 1 to 3 of a 7-day continuous infusion of cytosine arabinoside 100 mg/m2 per day; patients in remission received cyclic maintenance courses every 4 or every 8 weeks. The abstract names no control arm: the arm roles recorded here are a schema requirement.
Arms
  • Daunorubicin 30 mg/m2 (control): Daunorubicin 30 mg/m2 on days 1 to 3 plus cytarabine 100 mg/m2 per day by continuous infusion for 7 days
  • Daunorubicin 45 mg/m2 (experimental): Daunorubicin 45 mg/m2 on days 1 to 3 plus cytarabine 100 mg/m2 per day by continuous infusion for 7 days
  • Adriamycin 30 mg/m2 (experimental): Adriamycin (doxorubicin) 30 mg/m2 on days 1 to 3 plus cytarabine 100 mg/m2 per day by continuous infusion for 7 days

Complete remission ratePatients younger than 60 yearsprimaryYates 198272%59%The abstract states that daunorubicin 45 was significantly better than the other two arms in this age group; it gives no P value, confidence interval or number of patients per arm.
Complete remission ratePatients older than 60 yearsprimaryYates 198231%47%The abstract states that daunorubicin 30 was better than the other two arms in this age group, with a corresponding shift in induction mortality (no figures given); it gives no P value, confidence interval or number of patients per arm.
