ACTS-GC

Adjuvant chemotherapy for gastric cancer with S-1, an oral fluoropyrimidine

Phase 3NCT00152217

Population
Patients in Japan with stage II or III gastric cancer who underwent gastrectomy with extended (D2) lymph-node dissection.
Design
Randomised trial in Japan: surgery followed by adjuvant S-1 for one year (6-week cycles: 80 mg/m2/day for 4 weeks, 2 weeks off) or surgery only, after D2 gastrectomy for stage II or III gastric cancer. 529 and 530 patients were randomised between October 2001 and December 2004; the trial was stopped at the first interim analysis on the data monitoring committee's recommendation. The ClinicalTrials.gov record lists an estimated enrolment of 1000, which differs from the 529 plus 530 patients randomised in the paper.

Arms
  • Surgery only (control, n = 530): D2 gastrectomy with no adjuvant treatment
  • Surgery plus adjuvant S-1 (experimental, n = 529): S-1 for one year after D2 gastrectomy
