ABC-06
Second-line FOLFOX chemotherapy versus active symptom control for advanced biliary tract cancer (ABC-06): a phase 3, open-label, randomised, controlled trial
Phase 3N = 162NCT01926236
- Population
- Adults with histologically or cytologically verified locally advanced or metastatic biliary tract cancer (cholangiocarcinoma, gallbladder or ampullary carcinoma) with radiological progression on first-line cisplatin and gemcitabine and ECOG performance status 0 or 1
- Design
- Open-label randomised (1:1) phase 3 trial at 20 sites in the UK: active symptom control plus FOLFOX, or active symptom control alone. FOLFOX was given intravenously every 2 weeks for a maximum of 12 cycles. Randomisation used a minimisation algorithm with platinum sensitivity, serum albumin concentration and stage as stratification factors. The primary end point was overall survival in the intention-to-treat population. 162 patients were randomly assigned; median follow-up was 21.7 months.

The open full text states the primary analysis was a multivariable Cox regression adjusted for the stratification factors (platinum sensitivity, serum albumin concentration and disease stage). A post hoc sensitivity analysis using the stratification categories entered at randomisation gave an adjusted hazard ratio of 0.68 (95% CI 0.49 to 0.95; p=0.022). Median follow-up 21.7 months (IQR 17.2 to 30.8). |
